Abstract/Contents:"RNA-Seq analysis was conducted to evaluate differential expression in 2,4-D resistant and susceptible buckhorn plantain (Plantago lanceolata) 24 hours after 2,4-D treatment. In general, the susceptible biotype enriched numerous GO terms associated with parasitism, pathogen infection, and miscellaneous external biotic stress whereas such GO terms were not enriched in the resistant biotype. Two auxin associated GO categories were also enriched in susceptible, auxin efflux transmembrane transporter activity (GO: 0010329) and auxin transmembrane transporter activity (GO: 0080161), whereas no auxin-associated GO categories were enriched in resistant. Upregulated gene-level transcripts associated with plant defense response and parasitism were also observed in the susceptible but not in the resistant biotype. With respect to gene families associated with enhanced metabolic-based non-target site resistance, more gene level transcripts of cytochrome-P450s, glutathione S-transferases, and ABC transporters were upregulated in the susceptible biotype than the resistant biotype following 2,4-D treatment. Of the Aux/IAA repressors and TIR1/AFBs, no nonsynonymous mutations were observed in regions of auxin interaction. While it seems that 2,4-D resistant buckhorn plantain is not perceiving exogenously applied 2,4-D based on differential expression, the mechanism preventing perception remains unknown. Further research evaluating non-treated resistant and susceptible biotypes is currently being conducted."
