Abstract/Contents:"Lance nematodes (Hoplolaimus spp.) have become increasingly problematic on golf course putting greens across Virginia. Turfgrass professionals are monitoring plant parasitic nematode populations more frequently by sending routine samples into nematode assay labs for quantification. Previous research has demonstrated interlab variability in population estimates due to the use of different diagnostic methods, highlighting the need for a standard objective quantification method. The use of qPCR for quantitative estimates of various organisms is widely adopted for many diagnostic approaches, but is understudied for turfgrass nematode estimations. The objective of this research is to refine a qPCR technique for identification and quantification of lance nematodes from golf course putting greens. A standard assay of DNA extracted from 100 handpicked lance nematodes using a Qiagen Powersoil DNA Kit produced a cycle threshold range of 28.9-34.7. Lance nematode populations that were hand counted from plots (n=28) collected at Belmont Golf Course (BGC) in Richmond VA resulted in a cycle threshold range of 20.949-25.686. While still significant (p=.0352), the linear relationship between DNA quantity and hand counted lance nematodes for each plot was poor (r2= .16). The DNA of 250, 100, 50, and 20 handpicked lance nematodes was extracted using similar techniques to reduce human error and increase the strength of this relationship. The relationship of DNA quantity and hand counted lance nematodes for this reaction was stronger than the random samples from BGC (r2= .573, p=.0002). Our results from these studies suggest that extraction using a Qiagen Powersoil DNA Kit is a viable option for lance nematode identification and that these methods are reliable for estimating known quantities under controlled conditions. Future research will evaluate the linear relationship between DNA quantity and known lance nematode counts under these controlled conditions."
